皇仁書院校歌的旋律來自英國哈羅公學（Harrow School）的校歌《四十年來》，並與香港協恩中學的校歌旋律相同。英文校歌歌詞出自該校代理校長祈惠霖先生（Mr. William Kay）於1920年的手筆（祈惠霖先生在1934年6月接任成為英皇書院的校長）。而中文校歌歌詞則出自皇仁書院校友陳鈞潤先生的手筆，並刊載在校刊《黃龍報》內。雖然校歌有第二及第三段，但現時師生在任何場合唱校歌的時候均不會唱該兩段。
